What Is Foundayo® (Orforglipron)?

Your Titration Schedule: Starting Low, Going Slow

How Effective Is Foundayo®? The Clinical Data
Foundayo® was approved based on Eli Lilly's Phase 3 ATTAIN clinical trials, which enrolled more than 4,500 adults with obesity or overweight with weight-related conditions.
Here is what the data showed:
At the highest dose, participants lost an average of 12.4% of their body weight (approximately 27 lbs) over 72 weeks, compared to just 0.9% for those on placebo.
Across all trial participants (regardless of dose completion), the average weight loss was approximately 11.1% of body weight vs. 2.1% for placebo — a difference of about 25 lbs on average.
Participants also saw significant improvements in cardiovascular risk markers across all doses, including reductions in waist circumference, triglycerides, non-HDL cholesterol, and systolic blood pressure.
How does this compare to other medications?
It is important to note that there have been no head-to-head clinical trials comparing Foundayo® directly to any other weight loss medication, so direct comparisons should be interpreted carefully. In separate clinical trials (not head-to-head):
The Wegovy® pill (oral semaglutide) produced average weight loss of ~16% at the highest dose at 64 weeks.
The Wegovy® injection produced average weight loss of ~14.9% at 68 weeks.
Zepbound® (tirzepatide injection) produced average weight loss of approximately 21% at 72 weeks.
Experts note that in real-world use, ease of administration and adherence can significantly influence outcomes. Because Foundayo® has no food or timing restrictions and does not need to be refrigerated unlike the injectable GLP-1s, some patients may find it easier to take consistently, which can translate to real-world effectiveness. A Note for Our Female Patients: Oral Contraceptive Interaction

If you take oral birth control pills, Foundayo® may reduce their effectiveness. This is related to how GLP-1 medications affect the rate at which the stomach processes and absorbs other oral medications.
What this means for you:
Talk to your Phoenix NP provider before starting Foundayo® if you use oral contraceptives.
Your provider may recommend using an additional or alternative form of birth control for 30 days after starting Foundayo® and for 30 days after each dose increase.
Non-oral options (such as an IUD, patch, ring, or barrier methods) are not affected by this interaction.

Side Effects and Safety
